((Glycine)x4-Serine)x4 Linker
This linker consists of four repeats of four times glycine and one time serine. (Gly4Ser)4 linker is one of the most widely used flexible linker that can sustain the bioactivity of fusion proteins. It was used by the team FAU_Erlangen, 2019, to create a bispecific antibody targeting GPA33 on colorectal cancer cells and CD3 on T cells (BBa_K3117029, BBa_K3117026, BBa_K3117030).
Usage and Biology
Our flexible linker is composed of the small and non-polar amino acid glycine (G) and the polar amino acid serine (S). By adjusting the copy number, in this case four repeats of GGGGS, the length of this GS linker can be optimized to achieve appropriate separation of the proteins of interest (Chen et al., 2013).
